Display Fusion should be able to handle this, yup! You can create 2 monitor profiles, 1 for each setup. Attached to these profiles can have all the settings you would like to use for each setup.

I am considering buying a 38 inch monitor for my home office and would also like to get DisplayFusion to assist working with it. I am just slightly concerned what will happen at office, where I have 2 external 27 inch monitors.

Will DisplayFusion be able to automatically recognise and adapt accordingly when I am working with one big screen and with 2 smaller ones?

Changing the setup at office is unfortunately out of question, as I don't have a say to that. If life would get too awkward switching between these setups on a daily basis, I probably would just need to buy 2 screens to home office also.
