When trying to auto-update klite codecs, there is always an error message in their updater saying that DisplayFusion.exe is locking files preventing updates.

By killing the appropriate task the installation continues fine.

Is it possibly that displayfusion is locking something permanantly rather than locking during use?

Strange! It could be that it doesn't like that DisplayFusion is hooking whatever process it's trying to update. If you know which one it is, you can add it to a rule on the DisplayFusion Settings > Compatibility tab with the "Disable application hooks (this application)" option enabled in the rule.
